OPTIMASI NANOSTRUCTURED LIPID CARRIER LINESTRENOL DARI CAMPURAN PALM STEARIN DAN PALM KERNEL
Abstract
A nanostructured lipid carrier (NLC) was a lipid-based nanoparticle that was considered as a potential drug delivery carrier. Lipids as matrix of NLC can be found in natural sources, such as palm oil. Palm oil and its derivatives, such as Palm Stearin (PS) and Palm Kernel Oil (PKO), are widely available in Indonesia and can be used in the pharmaceutical sector. The main purpose of this study is to develop an optimized NLC formulation containing lynestrenol using a mixture of PS and PKO as a lipid matrix. NLC was prepared using high shear homogenization followed by the ultrasonication method. Process parameters in sonication such as duration, amplitude, and interval of sonication were optimized to obtain NLC with desirable characteristics. The optimal condition of sonication was then used to obtain NLC containing lynestrenol. The ratio of PS as solid liquid and PKO as liquid lipid used in the formulation of NLC containing lynestrenol was also optimized and evaluated for its particle size, polydispersity index (PDI), and zeta potential value. The results showed that the optimum condition of sonication was obtained with an amplitude of 20% for 4.5 minutes without intervals during the sonication process. Formula Fb-3 with ratio PS:PKO (6:4) and sonicated using obtained condition obtained desirable particle size (125.90 ± 3.24 nm), zeta potential value (-29.87 ± 1.94 mV), and PDI (0.21 ± 0.026). Therefore, the NLC obtained might be a potential delivery system for the transdermal route of lynestrenol.
ABSTRAK
Nanostructured lipid carrier (NLC) adalah salah satu nanopartikel berbasis lipid yang digunakan sebagai sistem pembawa obat. Lipid sebagai matriks penyusun NLC dapat ditemukan dari sumber alam, seperti dari minyak kelapa sawit. Minyak kelapa sawit dan turunannya, seperti fraksi minyak stearin (PS) dan fraksi inti kelapa sawit (PKO) banyak ditemukan di Indonesia dan dapat digunakan di sector farmasi. Penelitian ini bertujuan untuk optimasi formulasi NLC mengandung linestrenol, dimana NLC mengandung campuran fraksi PS dan PKO sebagai matriks lipid dari NLC. NLC dibuat dengan menggunakan metode high shear homogenization dan ultrasonikasi. Beberapa parameter sonikasi seperti durasi, amplitudo, dan interval sonikasi dioptimasi untuk mendapatkan NLC dengan karakteristik yang sesuai. Kondisi optimal dalam pembuatan NLC kemudian digunakan untuk menghasilkan NLC mengandung zat aktif linestrenol. Rasio dari PS sebagai lipid padat dan PKO sebagai lipid cair juga dioptimasi dan dilakukan evaluasi terhadap beberapa parameter seperti ukuran partikel, indeks polidispersitas, dan nilai zeta potensial. Hasil penelitian menunjukkan bahwa kondisi optimum sonikasi diperoleh dengan amplitudo 20% selama 4.5 menit tanpa adanya jeda selama proses sonikasi. Formula Fb-3 dengan rasio PS:PKO (6:4) yang disonikasi dengan kondisi optimum menghasilkan NLC dengan karakteristik yang diinginkan. Fb-3 menghasilkan ukuran partikel (125,90 ± 3,24 nm), zeta potensial (-29,87 ± 1,94 mV), dan PDI (0,21 ± 0,026). Sehingga NLC yang dihasilkan berpotensi untuk dijadikan sebagai sistem penghantar obat linestrenol melalui rute transdermal.
